Wide-field fluorescence microscopes

The Zeiss Axio Observer is an inverted microscope equipped with motorized XYZ stage, 5x, 20x, 40x, 60x and 100x Plan-Apo lenses, a Hamamatsu Flash 4.0 LT camera (2048x2048 pixels) and an environmental chamber for long-term live imaging. Fluorescence excitation is provided by the Zeiss Colibri 7 LED system, with filters suitable for almost every visible wavelength fluorophore. The system is operated using Zeiss's Zen Blue software. The Leica DMi6000 and DM6000 are automated microscopes outfitted with mercury light sources, filter cubes for basic epifluoresence microscopy, motorised XYZ stages, and Hamamatsu Orca R2 CCD cameras (1344x1024 pixels). Perfect for straightforward visualisation of fixed tissue and cell samples, and small microrganisms (yeast, chlamydomonas). The DMI6000 is an inverted system, the DM6000 is an upright system, but the microscopes are otherwise fundamentally similar instruments. The Nikon Eclipse TiE is a fully automated inverted microscope outfitted with two light sources for fluorescence imaging: a Heliphor LED-based source (405, 480, 555, 640 nm) to limit phototoxicity and a super bright, wide spectrum Sutter Lamda XL source with a collection of filter cubes for intracellular ion measurement. Two cameras are mounted on this sytem: a Photometrics Evolve 512 EMCCD for low light or single molecule applications and a Nikon DsRi2 CMOS (4908 x 3264 pixels) color camera. It is equipped with six lenses (4x, 10x, 20x, 40x, 60x, 100x), DIC optics, a motorized stage (including piezo Z control) and a gas- and temperature- controlled stagetop incubator. The system is operated using NIS-Elements software.
