This microscope is well suited for confocal fluorescent imaging of live cell/tissue preparations, viewing samples in culture dishes, culture plates (6 wells and greater), chamber slides that can be inverted and a variety of custom made (by user) chambers no larger than a standard culture plate. The live tissue chamber with atmosphere heater, stage heater, humidity and CO2 control permit long-term time-lapse imaging for a wide variety of applications. This microscope is also suitable for fixed cell/tissue imaging, using culture dishes or slides. Imaging through plastic dishes is limited to 5x, 10x and 20x lens. Higher magnification must be through coverslips on slides or Matek-type culture dishes. All slides MUST be sealed, clean and dry prior to placing on the stage. Imaging Modes: Laser scanning fluorescence and DIC (Nomarski) 3-D “Z-series” Time-series Tile scans (mosaics) Co-localization, FRET, FRAP, linear unmixing Multi-position FLUORESCENCE EXCITATION/EMISSION Long UV (405 nm for dyes such as DAPI, Hoechst 346, fluorogold) Blue/Green (458,488, 514nm for blue, green, yellow dyes, e.g., CFP,YFP,GFP, Cy-2, FITC-like dyes) Green/Red (561nm for Cy-3, Texas Red, or rhodamine-like dyes) Red/Far-Red (633 nm for far-red dyes like Cy-5) Emission bandwidth for all dyes/laser lines is continuously variable providing huge flexibility for collecting images of unusual dyes and for minimizing bleed-through between colors.
